The Communication was developed to address an important issue, namely the cardiovascular risks associated with nonsteroidal anti-inflammatory drugs (NSAIDs). This issue is particularly relevant to individuals taking NSAIDs, including Celebrex® and Celecoxib®, as they may have a higher risk of gastrointestinal (GI) adverse effects, especially in those taking higher doses (e.g., 400mg twice daily) or taking more than 1200mg in a 24-hour period. The increased risk of adverse reactions has been attributed to selective COX-2 inhibition or NSAID use, and the association with celecoxib is likely to be more complex in relation to GI side effects. This issue was recently addressed in a recent medical communication by Dr. J. Hsu and colleagues, in which they noted that the cardiovascular safety of celecoxib has been assessed in clinical trials, and that there has been no significant increase in the cardiovascular risk with celecoxib use in the long term. This prompted us to investigate the cardiovascular safety of Celebrex® Celecoxib®, which has been used since its patent life of production expired in 2006. We are also investigating the cardiovascular safety of the drug. The cardiovascular safety of Celebrex® Celecoxib® has been evaluated in more than 100 clinical trials. However, it is important to note that the cardiovascular safety of Celebrex® Celecoxib® is only applicable to those taking low doses of Celebrex® (400mg three times daily) and not to those taking higher doses of Celebrex® (200mg three times daily). This issue has not been addressed in our previous communication, which focused on a few cardiovascular risk factors.
A study by our group (Hsu & colleagues) looked at the cardiovascular safety of Celebrex® Celecoxib®, which has been used for several years now. The analysis found that the cardiovascular safety of Celebrex® Celecoxib® was not as well established as that of other NSAIDs. The current analysis focuses on patients with a diagnosis of gastro-esophageal reflux disease (GERD) who are taking high doses of celecoxib and who also have a history of ulcers, ulcerative colitis, or other GI ulcers. It is important to note that the cardiovascular safety of celecoxib has been assessed in clinical trials, and there have been no significant increases in the cardiovascular risk with celecoxib use in the long term. Although the cardiovascular safety of Celebrex® Celecoxib® has been assessed in more than 100 clinical trials, the overall cardiovascular safety of this drug has not been studied in more than 100 clinical trials.
